Interactome-wide analysis of the loss-of-function (LoF) effects of destabilizing mutations and their contribution to cancer-driving potential.

Scripts for structurally resolving the human protein interactome at the atomic level, mapping missense and nonsense mutations onto the structural interactomes, and distinguishing protein-folding destabilizing ("quasi-null") and protein-binding destabilizing ("edgetic") missense mutations. Such analyses allow for an interactome-wide analysis of the LoF effects of quasi-null and edgetic mutations and their contribution to cancer-driving potential.

Installation/Usage

Python 3 is required.

1. Clone or download this repository

2. Navigate to the downloaded CancerLoF folder

$ cd CancerLoF

3. Install the Python packages listed in requirements.txt

$ pip3 install -r requirements.txt

4. Follow the instructions and run the scripts listed in instructions.bash
