Hi:

It was an honor to read your paper on SBB in 2019: Woolet, J., Whitman, T., 2019. Pyrogenic organic matter effects on soil bacterial community composition. Soil Biology and Biochemistry.

What you describe in your method is the processing flow using DADA2. However, different studies cannot be directly merged due to the different 16S rRNA primer regions (Ling et al., 2022; Ramirez et al., 2018; Caporas et al., 2011; Yuan et al., 2020). These articles use closed references-based OTU picking protocol. So this confuses me whether it is possible to perform DADA2 directly, and if so, it is very convenient.
